The Fate and Ultrastructural Changes of Anabaena azollae Within the Debeloping Sporocarps of Azolla
Zheng Weiwen, Liu Lihua, Xiu Wenqiong
1998, 13(1): 1-8.
Abstract:
The fate and ultrastructural changes of Anabaena azollae entrapped into the developing sporocarps of Azolla pinnata were followed and characterized by laser confocal scanning image system (LCSIS) and transmission electron microscopy The entrapped vegetative Anabaena hormogonia tended to run parallelly to the long axis of the young sporocarps and underwent an active division, forming eventually an Anabaena colony, which occupied the entire indusial chamber and all the space available around the megasporangium or microsporangia Some of the vegetative cells initiated differentiation to akinetes while the opening of the chamber was closed At an intermediate stage of the sporocarp development, all the vegetative cells on the hormogonia gradually transformed into the akinete like chains Ultrastructurally the preakinetes underwent the cytoplasm reorganization and the membrane systems rearrangement The inclusions, such as carboxysomes, cyanophycin granules, ribosomes and thylakoids, varied in number, size, and their distribution in the cell along with the differentiation course of the akinete The similarities and differences in differentiation patterns and ultrastructural feature between the endophytic and free living cyanobacteria as well as their possible significance were discussed
Introduction of Maize Glutamine Synthetase (MGS)c DNA into Agrobacterium Tumefaciens
Tang Longfei, Liu Zhongzhu, Dong Kewen
1998, 13(1): 9-14.
Abstract:
The maize glutamine synthetase c DNA in Escherichia coli FDB213 was firstly subcloned into pBI121 vector by using sticky end ligation Seven of seventy two colonies tested (9.7%) were identified to be inserted with MGS c DNA The orientation of inserted c DNA fragments was measured by the double digestion of the subcloned plasmid DNAs with Hind III and Bstx I endonucleases Strains P9、P15、P16 and P17 have the insert from 5’ to 3’ (sense) and P2 from 3’ to 5’ (antisense) The subcloned plasmid DNA of P16 was further introduced into Agrobacterium tumefaciens 15955 by using direct gene transformation, freeze thaw method The transformed colonies were preliminarily selected by antibiotic screening method,50 u/ml kanamycin was supplied in LB medium, then the transformation was confirmed by Southern blot analysis A11 was identified to carry MGS c DNA fragment
Breeding and Selection of Mingguang 87s A Photosensitive Male Sterile of Indica Rice
Zhang Shougang, Xu Xuming, Zhuo Wei, Yang Tengbang, Huang Xianbo, Fan Zhujun
1998, 13(1): 15-20.
Abstract:
The keypoint to two line indica hybrid breeding is to successfully transfer PGMS genes directly or indirectly from Nongkeng 58s to indica varieties. An indica PGMS line,Mingguang 87s was newly obtained by crossing M 901s,a PGMS line derived from Nongkeng 58s which is highly stable sterility but lower outcrossing ability, with Jian 29s,a TGM line possesses high outcrossing performance. It became a completely male sterile under long day condition and restored to be fertile under short day condition. The critical temperature for fertility transformation is lower than 23 5℃.Under natural long day condition in summer,Mingguang 87s has a long (56 d ),complete (100%) and stable male sterility performance. In late autumn or winter (short day),its seed setting may reach as high as 74%.Type classification revealed that it belongs to indica subspecies. In addition,it possesses a favorable flowering performance and high combining ability.
A Newly Bred Rice Variety
Ye Xinfu, Zheng Jiuru, Lin Wenbin, Zhang Xin
1998, 13(1): 21-26.
Abstract:
Mancang 515”,a newly bred long duration(136 d) indicajaponica cross, possesses the traits of large panicles and heavy grain weight. It shows highly resistant to blast and is nonsensitive to photo temperature. It was registered and released in 1996 in Fujian province. In yield trials in 1994-1995 in northern Fujian,it averaged 8 17% more than Jinruo 6 and 8 26% more than Shanyougui 32.The research indicated that the breeding theoty about “ideal plant morphology combining with dominancy utilization” presented by Prof.Yang Shouren has been proved in the screening indica cline rice of indica japonica hybridization.
The Progress on Diagnosis and Detection of Citrus Huanglongbing
Tian Yanan, Ke Sui, Ke Chong
1998, 13(1): 27-35.
Abstract:
Diagnosis and detection are important aspect of citrus Huanglongbing research In the past, diagnosis of citrus Huanglongbing was mainly based on its sympomatology, namely, yellow shoot and mottle leaf During 1977-1978,the causative bacterium like organism (BLO) was found in sieve cells of diseased leaf under electron microscope After then, the electron microscope has been used as a major tool for diagnosis of citrus Huanglongbing disease An improvement came about when it was found that the BLO can be transmitted from citrus to periwinkle by dodder and partially purified by differential centrifugation, which made it possible to prepare antisera, both monoclonal and polyclonal antibodies, for detection and diagnosis of Huanglongbing BLO Recently, polymerase chain reaction (PCR) has been developed for detection and quanfitication of Huanglongbing BLO The PCR-based methods are highly sensitive and effective in detecting the presence of the BLO in diseased plant and insect vectors carrying the BLO Furthermore the BLO in the samples can be quantitated by quantitative PCR making it possible to compare the amounts of BLO in different samples.
Preparation of Crude Toxins of Ralstonia solanacearum in Sweet Potatoes and Influence of Heat、Ultraviolet Ray、Enzyme to the Biological Activity of Crude Toxins
Zhong Zangwen, Lu Tong, Li Benjin, Kang Yumei, Chen Qinghe
1998, 13(1): 36-40.
Abstract:
The strains with pathogenicity of Ralstonia solanacearum isolated from sweet potatoes were grown in PYG medium at 28-30℃ for 48-72 hours After sterile centrifugation (2000 r/min ),the culture fluids were filtered through 0 45 μm Millipore filters The crude toxins (metabolite) were obtained The crude toxins of Ralstonia solanacearum in sweet potatoes is sensitive to thermal treatment and it’s biological activity decreased separately 0%、28 7%、100% at 50℃、 70℃ and 100℃ waterbath for one hour The crude toxins is not sensitive to ultraviolet ray and the biological activity is unchanged under ultraviolet light for 30min、60min and 120min The crude toxins is sensitive to Pronase and lysozyme, the biological activity was destroyed and the toxicity was lost while adding papayain (4-16 u/ml) or trypsin (4-16 u/ml) to the crude toxins It’s biological activity decreased 91 6%-92 0% while adding lysozyme(15000-60000 u/ml) to the crude toxins.
