Molecular Epidemiology of Muscovy Duck Reovirus

Abstract:
The primers were designed to amplify the S1 and S4 gene according to the muscovy duck reovirus(MDRV) sequencein Genbank. S1 and S4 gene of MDRV strains isolated in China were cloned and then sequenced and analyzed. The results of showed S4 and S1 gene was 992 bp and 819 bp in length, respectively; the nucleotide sequence of S1 segment shared 92.2%-96.7% identity in China strains, but 92.2%-96.7% identity with France strain, 89026; and 76%-78% identity with ARV strain, S1133. The nucleotide suquence of S4 segment shared 95.2%-99.3% idnentity in China stains, but 92.3%-94.5% identity with France strain, 89026, and 21.2%-21.5% identity with ARV strain,S1133. Phylogenetic analysis demonstrated that MDRV was quite different from ARV. The Ten MDRV strains isolated in China has no distinct difference, but they were different from 89026 (isolated in France). The study provide the basic research on the prevention with muscovy duck reovirusis.
The Growth Advantage of the Pectoral Fin Cells of Anguilla Anguilla Under Different Culture Conditions

Abstract:
For establish a cell model of Anguilla Anguilla which can be used for isolation and identification of Anguilla Anguilla virus, the pectoral fin cells of Anguilla Anguilla (the 21st generation) were used in this study. The pectoral fin cells of Anguilla Anguilla were cultured in artificial medium in vitro to explore the optimum conditions. The pectoral fin cells of Anguilla Anguilla were cultured under different mediums (L-15, RPMI-1640, and DMEM), different serums (BS, FBS), and different serum concentrations (5%, 10%, 15%, and 20%). In different time points, the cells were collected and counted, and then the growth curves of the cells were drawn. The effects of the different conditions on the proliferation of the pectoral fin cells were evaluated by the growth curves. The results showed the L-15 was the optimum medium,while RPMI-1640 took the second place and DMEM was the poorest; FBS was better than BS, while 10%FBS was the best concentration.
The Effect of Dietary Crude Protein and Digestible Lysine on Growth Performance and Nitrogen Excretion in Growing Pig

Abstract:
90 DLY cross bred pigs (32.0?1.5 kg) were randomly allocated into 5 groups, 3 replicates per group, 6 pigs in each replicate to investigate the effects of dietary protein and lysine on growth performance and nitrogen excretion in growing pigs. 2?2 factors were designed. Control group was fed with diet of 17.47% crude protein (CP) and 0.80% digestible Lys ( Dly ). 4 treatment groups was fed with diet of 15.46%, 13.49% CP and 0.80%, 0.85% Dly, respectively. The test duration was lasted for 50 days. The results showed that the treatment fed with diet of 15.46% CP and 0.80% Dly has not effect on the growth performance in growing pigs in comparison with others (P>0.05),but compared to the control, urinary nitrogen was significant decreased (PPPP>0.05) compared to the others, urinary and feces nitrogen and total nitrogen was significant decreased (PP>0.05) but at the same time nitrogen excretion is significant decreased in comparison with the diet of 17.47% CP and 0.80% Dly (P<0.05).
Acute Toxicity Tests of Four Heavy Metal Salts to Juvenile Snail of Babylonia lutosa

Abstract:
The acute toxicity of four heavy metals to juvenile snails of Babylonia lutosa were studied at water temperature 16-23℃, salinity 30 and pH 8.0. The results showed that the LC50 values of CuSO4·5H2O treatment for 24h, 48h, 72h and 96h were 2.596, 1.037, 0.690 and 0.400 mg·L-1, respectively. The LC50 values of ZnSO4·7H2O treatment for 24 h, 48 h, 72 h and 96h were 16.384, 4.738, 3.225 and 2.721 mg·L-1 respectively. The LC50 values of CdCl2 treatment for 24 h, 48 h, 72 h and 96h were 17.286, 7.340, 3.799 and 2.361 mg·L-1 respectively. The LC50 values of Pb(NO3)2 treatment for 24h, 48h, 72h and 96h were 650.640, 410.595, 262.342 and 141.525mg·L-1 respectively. The safe concentration of four heavy meals to juvenile snails of B. lutosa were 0.040, 0.272, 0.236 and 14.153 mg·L-1 respectively.The safe concentration of four heavy metals ions to juvenile snails of B.lutosa were 0.010,0.062,0.148 and 8.501 mg·L-1 respectively.The toxicity was in the order of Cu2+>Zn2+>Cd2+>Pb2+.
The Artificial Infection of Duck's Cecum Enlargement and Necrosis Syndrome

Abstract:
For the etiological researching of Duck's Cecum Enlargement and Necrosis Syndrome (DCENS), 38 20-days old ducks were fed with organ mixture from duck infected with DCENS, loach from the onset area and metacercariae isolated from loach respectively. Those fed with loachs and with 500 metacercariae showed typical symptoms of DCENS; but those fed with 100 metacercariae showed only mild symptoms and cecal lesion. The average number of Cyathocotyle caecumalis sp.nov isolated from the duck cecum of the group fed with loachs, 500 metacercariae and 100 metacercariae were 39, 61.7and 11.8, and eggs were checked out from all the three groups; while the group fed with organ mixture showed none symptom and Cyathocotyle caecumalis sp.nov detection. The results suggested that C. caecumalis sp.nov is the only pathogen of DSCEN. The loach from the paddy field of the onset area is the second intermediate host of C. caecumalis sp.nov.

Effects of Different Sowing Dates on Plant type and Yield Traits of Hybrid Rice Variety Yiyou 673

Abstract:
Interval sowing experiment was executed with Yiyou 673 as testing material to explore the effects of different sowing time on the growth period, plant and leaf morphology and yield components. The results showed that the growth period of rice was shorten, the number of main stem leaves was reduced and the plant height was decreased with the delayed sowing date. At the same time, the number of effective panicles was reduced, while the seed setting rate was increased, and the grains per spike were firstly ascended and then declined gradually. Optimum sowing time for Yiyou 673 had been confirmed on 20th May, it had good plant and leaf morphology, big panicle with more grains and the highest yield. Yiyou 673 could gain higher yield for sowing on 1st May, but it would have lodging risk because of over high plant height in this period. Therefore, we should pay attention to strengthen the scientific cultivation of fertilizer management, drain paddy fields timely, combine the promotion and control, develop strong roots and stalks so as to enhance the capacity of lodging.
Correlation Between Agronomic Characteristics and Productivity of New Peanut Varieties from South China

Abstract:
Nine newly bred peanut varieties in south China were used in the study. Correlation analysis showed that the percentages of plump pods and shelling were very significantly correlated with the plant productivity. The stem height, branch length and pod branches of a peanut plant were positively, but not significantly, correlated to its productivity. The total number of branches and 100-pod weight were negatively correlated with the plant productivity. Path analysis showed that the direct path coefficients between the yield traits and plant productivity ranging from high to low were as follows: shelling percentage, branch length, 100-pod weight, total branch number, plump pod percentage, pod branches and stem height. In breeding high-yield peanut varieties, selection of the traits, such as pod number, plump pods, shelling and pod size should be important. While, other agronomic characteristics could be secondary for consideration.
Breeding and Cultivation of New Peanut Variety Fuhua7 with High Protein and High O/L Ratio

Abstract:
A new peanut variety Fuhua7 was bred from the cross between zhanyou41 and 9817-36-2 by using hybrid pedigree method in the spring 2002. Fuhua7 took part in spring peanut variety regional tests in Jiangxi province in 2009 to 2010.Its average pod yield and seed yield were 4680.2 kg穐m-2 and 3234.2 kg穐m-2, and increased by 5.62% and 6.94% compared to Shanyou523, respectively. The crude fat content, protein content and oleic acid/linoleic acid value of the seed was 52.02%, 30.19% and 1.35, respectively. The variety had high drought resistance, medium flooding and lodging tolerance, lighter leaf spot, rust resistance, bacterial wilt susceptivity, higher and more stable yield. The whole growth period was about 123 days. It was released in 2011 in Jiangxi province.

Cloning and Structural Analysis of Phosphoglycerate Mutas Gene and Its Expression Levels in Homokaryon and Heterokaryon of Volvariella volvacea

Abstract:
Two single spore isolates named PYd21 and PYd15 respectively and their hybrid heterokaryon named H15-21 were studied as materials in this paper. Using sequencing data of genome, transcriptome and digital gene expression profiles of Volvariella volvacea, Phosphoglycerate mutas(PGAM) were cloned and its structure and expression were studied. Alignment of PGAM genes from PYd21 and PYd15 revealed that both sequences to be identical. Structural analysis of PGAM gene revealed that it consisted of 2118bp with an open reading frame(ORF) of 1611bp,encoding a polypeptide of 536 amino acids. PGAM gene contained 10 exons and 9 introns. One type of alternative splicing and 6 alternative splicing sites were identified during RNA processing. TPM(Transcript Per Million Clean Tags) values for PYd21,PYd15 and H15-21 were 35.29,127.91 and 302.89 respectively, which had significant positive correlation to the growth rate of mycelium. The expression levels of PGAM gene in PYd21,PYd15 and H15-21 indicated that two differently type of nucleus in the cell of H1521 interacted each other which affected expression level of PGAM gene in H15-21. The expression levels of PGAM gene were confirmed by real-time quantitative PCR.

Abstract:
Through field investigation, seven broccoli cytoplasmic male sterile materials were selected out of 19 crossbreed materials of Japanese broccoli. Six cabbage cytoplasmic male sterile lines were obtained by crossing the male sterile materials with cabbage inbred lines as recurrent parents; the sterile lines showed excellent general characteristics with high combining ability, both of sterile plant rates and sterility reached 100%. Five excellent hybridized combinations were obtained by crossing with the male sterile lines. Xiahua No.2 is a variety that developed through the field screening.
Genetic Relationship Analysis of 24 White-fleshed Loquat (Eriobotrya japonica) Germplasms by ISSR

Abstract:
Genetic relationship of 24 white flesh loquat germplasms was studied by ISSR technique. 16 primers which could produce clear and polymorphic bands were screened out from 89 ISSR random primers. 195 locis were detected, of which 153 were polymorphic and the polymorphic percentage was 78.46%. As analyzed by NTSYS-PC2.0, the genetic similarity ranged from 0.55-0.89.24 white-flesh loquat germplasms were grouped into three sub-cluster when the similar coefficient was 0.72. Rich genetic diversity of white-flesh loquat germplasm resources was detected at DNA level. in which relationship were not so obvious correlation with geographic origin. Most germplasm resources which came from the same geographic origin had close relationship, But some germplasm resources with different source regions were clustered in a group obviously. In addition, during the reserch we also found that 7 special bands could be reference mark of 6 Loquat cultivars identity. Some specific ISSR markers could identify Guifei, Xinbai No.1 and Xinbai No.8.
Effects of Exogenous Abscisic Acid on Growth and several Physiological Characteristics of Broccoli under Cold Stress

Abstract:
The growth situation and activity variation of SOD and POD of different broccoli cultivars under cold stress were evaluated. After that, the relaxation effect of exogenous ABA on plant growth was studied by using broccoli cultivar ‘Cuilv’ (CL) as test material. The results showed that: (1) Exposure of broccolis to cold stress resulted in inhibition on plant height and stem diameter. Meanwhile, the activities of SOD and POD and soluble sugar content of the broccolis decreased significantly. However, different cultivars showed different responses to cold stress. The cultivar CL and MTL were greatly suppressed by cold stress, while cultivar YX and AU showed less influence. (2) ABA at 100 mg·L-1 improved plant height and stem diameter of CL significantly. The SOD and POD activities of plants treated by 50, 100 and 150 mg·L-1 of ABA were significantly higher than that of control. In addition, 100 mg·L-1 ABA increased mostly the activities of SOD and POD among all treatments. (3) Exogenous ABA significantly enhanced the soluble sugar content of CL. After 15 days of cold treatment followed by 15 days of recovery treatment, the soluble sugar contents of CL plants treated by 50, 100 and 150 mg·L-1 ABA were significantly higher than that of control and maintained at a high level.
Correlation Between Natural Branch Characteristics and Quality of White-flesh Loquat Fruits

Abstract:
Under natural conditions, correlation between the characteristics of fruiting branches and quality of Guifei loquat fruits were studied.The results indicated that the leaf number and branch length significantly affected the panicle weight.They might serve as a major index for spring shoots.For the summer shoots, it's the branch length affected markedly on the fruit quality, while the number of shoots significantly affected the fruit setting.Under the natural conditions, to produce high quality loquat, the followings were found to be desirable for the spring shoots: diameter > 0.9 cm, length > 20 cm, and 15-19 leaves per branch, while for the summer shoots: diameter > 0.7 cm, length > 30 cm, and more than 20 leaves on a branch.

Analysis of Microbe and Nutrient Characteristics of Cold-waterlogged Paddy Soil in Fujian Province

Abstract:
The microorganism population and nutrient contents of 14 soil samle collected from different paddy fields in 6 counties of Fujian Province were examined based on classical methods. The differences of fundamental natures between cold-waterlogged paddy soil (CWPS) distinguished and non-cold-waterlogged paddy soil (NCWPS) were also investigated. The results showed that (1) significant differences in some soil properties were detected between CWPS and NCWPS, including organic matter (OM) content, available P (AP) content and fungus number in the 0-20 cm soil layer. Compared with NCWPS, CWPS had more OM content, less AP content and fungus number in the 0-20 cm soil layer; (2) there are significant and positive correlations between bacterium, fungus and antinomycete number in NCWPS, but not in CWPS; (3) fungus number were negatively correlated with organic matter content and positively correlated with available P content, and antinomycete number were positively correlated with available P content. The number of fungus and antinomycete in soil significantly decreased with increase of soil depth. Compared with bacterium, fungus and antinomycete were easily influenced by soil physicochemical factors.

Identification of Cry-type Gene of Bacillus thuringiensis Strain LSZ 9408

Abstract:
PCR and SDS-PAGE were used to identify the cry-type gene of Bacillus thuringiensis (Bt) strain LSZ 9408. PCR analysis showed that Bt LSZ 9408 strain contained cry1 and cry2 genes. Five pairs of general primers-Un1, Un2, Un3, Un4 and Un7/8 were used to amplify PCR, and then the amplification results were detected by SDS-PAGE. Primes Un1 and Un2 formed about 270 bp and 700 bp DNA fragments on agarose gel electrophoresis, while none PCR products was found with the other three pairs of primers. The results of SDS-PAGE also indicated that Bt LSZ 9408 possessed cry1 and cry2 genes, which coded 130kD and 65kD ICPs.
Growth Fitness in Fermentation Process of FJAT-T8, an Avirulent Ralstonia Aolanacearum Used as Plant Vaccine Against Crop Bacterial wilt Disease

Abstract:
The paper dealt with the fermentation technique of an avirulent Ralstonia solanacearum strain FJAT-T8, which was used as plant vaccine against crop bacterial wilt disease. The fermentation process of FJAT-T8 was investigated by determination of on-line and off-line parameters in 50L automatic fermentation container. In order to study the growth fitness of FJAT-T8, the parameters with significantly variation during fermentation process were served as indexes and analyzed with cluster analysis. The multiple linear regression and correlation analysis were also used to analysis the parameter dynamic model and relationship between fermentation factors. Besides, tracing observation of cell morphology was conducted with electron microscope. The results showed that the cell growth cycle of FJAT-T8 could be divided into four periods: 0-16h (adaptation period), 24h (logarithmic period), 32-72h (stationary period) and 80-96h (decline period). The comprehensive evaluation indexes for the best fermentation quality were: fermentation time of 40-48h, fermentation pH of 8.7-8.8 and the dissolved oxygen maintaining steady. The study would improve the fermentation quality, formation of product quality standards and factory production of plant vaccine strain T8.

GC/MS Analysis on Acetone Extracts from Microbial Fresh-Keeping Agent for Fruit longan

Abstract:
Acetone extracts of mrcrobial Longan fresh-keeping agent were analyzed by GC-MS. Thirteen-eight compounds were identified. Five compounds with the match degree above 90%, including 2-Furanmethanol; 2-Furancarboxaldehyde,5-methyl-; Malto; 1,4∶3,6-Dianhydro- alpha -d-glucopyranose; Pyrrolopyrazine-1,4-dione, hexahydro-. The results showed that the main components were Pyrrolopyrazine-1,4-dione, hexahydro- and 2-Furancarboxaldehyde, 5-methyl-, with relative amount of 37.52% and 4.97%.2-Furancarboxaldehyde, 5-methyl- and Maltol were the aromatic compounds of Microbial Longan Fresh-keeping Angent.
Studying Storage Stability of Spray Drying Powder from Blanching Water of Stropharia rugoso-annulata

Abstract:
The spray drying powder of blanching water for Stropharia rugoso-annulata falow was studied to understand its changes in physical characteristics,hygroscopicity,free radical scavenging activity during storage period. The results showed that, the lower of storage temperature for spray drying powder, the higher in scavenging efficiency of it for ·OH free radical, and the longer in its half-lost time. The low temperature preservation was suggested for spray drying powder and it should be under the 20℃. With same storage temperature, its half-lost time in darkness was longer than it exposed in light and the higher the relative humidity, the faster the moisture absorption was. According to the difference of moisture content between spray drying powder and packing standard, the release time before packing of the spary drying powder was suggested 0.34 hour even under 92.31% of relative moisture. The results indicated that stropharia rugoso-annulata blanching powder by spray drying revealed a good stability while stored under low temperature、low humidity and dark conditions, especially in avoiding moisture regain during the storage period.

Status and Prospect of the Research on the Breeding of Hybrid Rice in Fujian Province

Abstract:
According to relevant literature, it was counted that the information of 184 hybrid rice varieties which had been registered in Fujian Province from 1991 to 2010. The achievements of hybrid rice breeding of Fujian Province in recent 20 years were reviewed. The parents of 184 registered hybrid rice varieties were analyzed in this paper. Results showed that these varieties were mainly bred by scientific and technical personnel of Fujian Province, making up 85.87% of the total. The utilization of male sterile lines in the breeding of hybrid rice was relatively simple during the 8th and 9th "Five-Year Plan". But, more male sterile lines were used during the 10th and 11th "Five-Year Plan", such as "Ⅱ-32A", "LongtepuA", "Zhenshan97A","Gang46A", "D62A" and "SE21S". Higher frequency applications restorer lines had "Shuhui527", "Minghui86", "Minghui63", "Fuhui673" and "Minhui3301" from 1991 to 2010. However, the disadvantages of hybrid rice breeding in Fujian were fewer of varieties which possessed high quality or good comprehensive characters of high yield, resistant and quality, and lack of varieties which possessed resistance to multiple pests and diseases, and suitability of simplified cultivation. Based on the above analysis, the strategy of hybrid rice breeding was proposed in this paper.
