Abstract:
Objective Sequence of the promoter region of (S)- to (R)-reticuline gene (STORR) involving biosynthesis of Papaver somniferum L. secondary metabolites, benzylisoquinoline alkaloids (BIAs) including morphine, was determined for further studies.
Method Using leaves of opium poppy plant at seedling stage, the known STORR upstream promoter sequence was cloned by Tail-PCR. Core promoter region and CpG island were predicted, and cis-acting elements analyzed. STORR expressions in the roots, leaves, and capsules collected at 0, 3, 6, 24, and 48h of the plants at capsule expansion stage treated by methyl jasmonate (MeJA) were determined using qRT-PCR.
Result A 4,156bp effective sequence upstream of STORR was successfully amplified. The promoter was predicted to have 12 core regions without CpG island and contained 246 cis-acting elements. Plants treated by MeJA were induced to express the gene in the roots, leaves, and capsules.
Conclusion The upstream promoter region of STORR was successfully cloned by Tail-PCR with potential core regions predicted and no CpG islands found. The cis-acting elements covered pathways of initiation, light response, hormone signal, and growth and development. STORR expressions exhibited in the roots, leaves, and capsules of the MeJA-treated opium poppy plants.
