Comparison of Biological Characteristics of Two Different Disease-type Duck Reovirus(MDRV and NDRV)

Abstract:
Muscovy duck reovirus(MDRV) and Novel duck reovirus(NDRV),which belong to Orthoreovirus,are important pathogens of waterfowl.This study was conducted to compare the biological characteristics of MDRV and NDRV.Several methods were undertaken such as ultrathin sections observation,cross virus-neutralization test,the pathogenicity to avian embryos and cell lines,comparison of genome segment profiles and the characteristics analysis of S3 gene.The results of ultrathin sections observation showed that MDRV and NDRV viral particles were spherical and the diameter was both about 70 nm.MDRV scattered in MDEF cytoplasm,whereas NDRV particles arranged in crystal lattice.Relatedness(R) values showed that the antigen of the two viruses were generally different.MDRV′s CPE feature was cell rounding,while which of NDRV was cell fusion.NDRV induced the death of 9-day-old chicken embryos,whereas MDRV did not.The migration profiles of NDRV and MDRV genome segments were different.The S3 genes of the two viruses were at different branches of molecular phylogenetic tree.In conclusion,the biological characteristics of MDRV and NDRV were obviously different.
Cloning and Polymorphism Analysis of VIPR-1 Gene in Black Muscovy Duck

Abstract:
As a critical candidate gene of poultry broodiness,the length of 1 864 bp(target 1) and 1 673 bp(target 2) gene fragments of the vasoactive intestinal peptide-1 gene were amplified with polymerase chain reaction method from the genomic DNAs Abstracted from the blood of black Muscovy duck.The results of sequence analysis demonstrated that the target fragment 1 included the complete sequence of the 5th exon(101 bp),the 5th intron(1 630 bp) and the 6th exon(133 bp).The target fragment 2 contained the complete sequence of the 12th exons(42 bp),the 12th introns(1 455 bp) and partial sequence of the 13th exons(176 bp).Another four pairs of specific primers(P1-P8) were designed according to the two sequences fragments,respectively.The results showed that the target fragment 1 had C/T mutation at position 318 bp and 454 bp,A/G mutation at 547 bp,A/T mutation at 923 bp;the target fragment 2 had C/T mutation at 89 bp and 944 bp,A/G mutation at 662 bp and 1 031 bp,A/C mutation at 1 334 bp.The characterization and mutations discovery with the VIPR-1 gene provided the basic evidences for the further study on the association of its polymorphisms with broodiness trait with the black Muscovy ducks.

Abstract:
A novel speed LAMP assay for GPV was explored with Loop-mediated amplification(LAMP).With two pairs of identify six positions in VP3 gene,a LAMP assay was developed for the rapid detection GPV.The results indicated that GPV was detected by LAMP assay at 65℃ as early as 50 min of incubation in a simple water bath.The detection limit of the LAMP assay was 150 pg·μL-1 of GPV genome.And no amplification was found with the samples of other viruses.A mixture of calcein and Mn2+ was used as fluorescent reagent to make the result of LAMP visible.Therefore,the simple and fast method is a potential useful technique for GPV detection in the field.
Genetic Characteristics of ORF1 Gene of Porcine Torque Teno Virus Genogroup 2 in Fujian

Abstract:
The ORF1 Gene of porcine torque teno virus genogroup 2 was amplified by PCR method from the piglets sera genome association with post-weaning multisystem wasting syndrome from Fujian,the target product was cloned and sequenced.The results demonstrated that the porcine torque teno virus genogroup 2 ORF1 gene was 1875 bp in length,coding an open reading frame(ORF) with 624 amino acids.The nucleotide sequence and the amino acid sequence deduced from the gene were analyzed by the bioinformatics software.Compared with the porcine TTV2 ORF1 genes download from GenBank,the sequenced gene shared the highest homogeneity with FJ/China/2010/TTV2/2(GenBank accession number JF937656) strain at 98.8%,but shared lower homogeneity with SC strain(GenBank accession number HQ204188) strain only at 83.5%.According to the results of the phylogenetic relationship,the porcine torque teno virus had three subbranches,the Fujian isolates had two different branches,which means that there had more than two TTV2 sub-genogroups in Fujian,China.
Establishment of an Indirect ELISA for Detecting Egg Yolk Antibody Against Chicken Flavivirus

Abstract:
A novel Flavivirus infection,which mainly caused egg-dropping in ducks and chickens,has spread throughout China since 2010.To carry out the serological surveillance in chickens with minimum stress,an indirect enzyme-linked immuosorbent assay(ELISA) for detecting egg yolk antibody against Flavivirus in layer chickens has been developed by coating plate with chicken-origin Flavivirus FQ-C1 isolate.The optimal working concentration of antigen was 5.7 ng ·μL-1,the working concentration of egg yolk antibody was 1∶160 dilution and that of HRP-labeled goat anti-chicken IgG was 1∶3 200.After a color reaction of 10 minutes with Tetramethylbenzidine(TMB) liquid substrate,a threshold of positive/negative(P/N) ≥ 2.1 was determined as positive standard for the ELISA.The results indicated that the ELISA established by this research provided a high sensitivity and specificity test,which was suitable for the detection of antibodies against Flavivirus in immune laying hens.

Abstract:
The aim of this study was fractionate and analysis of feather peptide for feed.5 components contented in feather peptide meal were separated by AKTA purifier 100 with Sephacryl S-100.The molecular weight of them were as followed: >2 000 000,6 456.54,758.58,398.11,186.21.Resultsof fractionation showed the fifth component were Asp,Thr,Ser,Glu.The fourth component were dipeptide or tripeptide,which content Asp,Thr,Ser,Gly,Ala.Amino acid composition of the third component,six peptide or seven peptide,were as followed: Asp,Thr,Ser,Glu,Gly,Ala.Peptide length of component 2,whose amino acid composition is the same as component 1,was close to antibacterial peptide.All these showed feather peptide meal was one kind of potential protein source for feed as it was rich in small peptides.

Abstract:
The genetic diversity and phylogenetic relationship of 94 yam germplasm was analyzed by ISSR.24 primers were selected from 100 primers and 352 clear strips were amplified.344 clear strips were polymorphic bands and the percentage of polymorphic bands was 97.7%.Genetic cluster analysis indiated that 94 yam germplasm were divided into 4 groups and the genetic distances were Dioscorea(0.053 8-0.088 4),Dioscorea alata(0.035 3-0.068 2),Dioscorea fordii(0.053 3-0.138 2) and Dioscorea persimilis(0.054 2-0.126 6).Based on the genetic similarity and the population,results suggested that Dioscorea alata group had the closest phylogenetic relationship with Dioscorea fordii group,closer with Dioscorea group and farther with Dioscorea persimilis group.
Effect of Potassium Fertilizing on Yield of Fall Sweet-potato

Abstract:
Relationship between applying K fertilizer and the yield of sweet potato was studied with variety of Xicheng Potato 007.The results showed that: K fertilizer improved the assimilative capacity of the sweet potato,and increased the dry matter accumulation while the plants partly inhibited in excessive growth of the stems and leaves.K fertilizer enhanced the plant capability in transporting assimilate to underground tuber,by which reducing the value of T/R and increasing economic production.Root dry rate and stem diameter were related with the species characteristics of sweet potato instead of K fertilizer amount.Reducing K fertilizer amount increased the portion in large-and medium-sized sweet potato,the length of the longest stem and the number of base branches.Comparing to control,the dry matter content was decreased by applying K fertilizer.The chlorophyll content of plants was higher in those treated with N and P fertilizer than with K fertilizer and without fertilization.For a low K level soil,the optimal amount of K fertilizer was 253.05-262.46 kg·hm-2 for producing high yield of dried sweet potato and starch,and was 316.05-321.59 kg·hm-2 for producing high yield of fresh potato tuber yield.There was no difference observed between low K and high K levels on dry matter production,although K fertilizer improved fresh yield effectively.

Abstract:
A new peanut variety Puhua4 has been bred from Yueyou7 and Jinhua1022 by Putian Institute of Agricultural Sciences.The new variety is characterized in high and stable yield,diseases and lodge resistance.The variety performed good in agricultural properties of all growing stages,such as relatively centered for blooming period,great for leaf area index,net assimilation rate and dry matter accumulation,capable of podding with big pod and kernel.It is suitable for growing in both dry land and paddy field in Fujian.The variety was certificated and released by Fujian Crop Variety Approval Committee in 2012.
Preliminary Study on Regional Suitability of Pear Varieties with Differences in Maturity

Abstract:
To adjust and optimize the variety structure of pear production in Fujian,an adaptive experiment was conducted in three culture regions with seven new varieties.The varieties were introduced from other provinces or abroad with differences in maturity stages.The results showed that 'Cuiyu' revealed advantages in fruit quality for commercializing,early maturity,ratio of fruit setting and yield,and with a good prospects for popularization and application.'Chuxialv' presented the potential to be a variety of complement to early-maturing pear.Other pear varieties showed no significant advantage in the aspects of comprehensive cultivation and economic characters compared to 'Cuiguan' and 'Huanghua',which should be careful for developing as cultivars.
Effect of Controlled Release Fertilizer on the Yield and Quality of Banana and Nutrient Use Efficiency

Abstract:
A field experiment was conducted to study the effect of controlled release fertilizer CRF on banana in Zhangzhou of Fujian province.The results showed that the yield of banana after application of CRF was increased by 6.18%(2 204.7 kg·hm-2) and the total soluble solids and edible part ratio were also increased compared with the normal NPK fertilizers at medium soil fertility.Meanwhile,the uptakes of N,P and K were 277.44,33.43 and 974.51 kg·hm-2,respectively,and the utilization efficiency of N,P and K were 15.1%,17.4% and 34.7%.Our results suggest that,compared with the normal N,P and K fertilizers,the use efficiency of CRF were increased by 42.7%,1.2% and 10.4%,respectively.

Abstract:
Embryonic tips from 'Mindou No.5' were used in testing the effect of NaClO on sterilization and homomycin B on screening stress for inducing of adventitious buds and root growing.The result showed that the optimal effect on sterilization was 10% NaClO for 10 min;the screening stress of hygromycin B was 7 mg·L-1 at elongation stage of buds from embryonic tips.Comparing culture mediums,the rooting culture medium presented better in root growing without 6-BA,the medium consisting of 1/2 MS salt+1/2 B5 organic matter +3% sucrose+0.56% agar+1.0 mg·L-1 IBA,pH 5.8.

Abstract:
Cut-flowers Eustoma grandiforum were steeped by different concentrations of salicylic acid.The changes of fresh weight,flower diameter,water balance values,relative conductivity,soluble protein content,praline content and soluble sugar content were investigated.The results showed that suitable concentration of salicylic acid could prolong fresh day,increase fresh weigh,maintain water balance,delay the degradation of soluble protein,lower the relative membranes permeability and reduce the content of proline.Our results suggested that 60 mg·L-1 salicylic acid was the best fresh-keeping effect.
Investigation of Organic Chlorine Residues in the Soil of Tea Garden

Abstract:
The organochlorine pesticide residues(HCHs and DDTs) in the organic tea garden soil in Fujian were investigated by GC analysis.The results showed that HCHs and DDTs were detected at different levels among 21 tea garden soils.In addition,the residues total of HCHs was 0.10 μg·kg-1 with β-666 and α-666 as the most residues;the residues total of DDTs was 0.18 μg·kg-1 with pp-dde,pp-ddd and op-ddt as the most residues.Further,the HCHs and DDTs residues in high-altitude tea gardens were more than low-altitude tea gardens significantly and correlated with tea ages negatively,and single pollution index and synthetic pollution indices suggest that BHC and DDT residues are not exceeded and the soil is suitable for tea cultivation.
Optimizing Nutritional Value of Wheat Bran through Pleurotus geesteranus Cultivation on a Selenium-enriched Medium

Abstract:
Taking advantage of the metabolic transformation on a selenium(Se)-enriched wheat bran medium by Pleurotus geesteranus,the nutritional value of the resultant dietary fiber powder could be substantially improved.The average content of biological organic Se content in the wheat bran powder reached 1.63 mg/kg with an added 3 μg·g-1 of sodium selenite that is within the safe threshold of Se nutrition metabolization.The amount was more than 56 times of the Se content in wheat bran,indicating an average Se conversion efficiency of approximately 86%.Aside from Se,the dietary fiber,total protein,total soluble sugars and proteins in the treatment powder did not differ significantly from the control.The result suggested that the cultivation of P.geesteranus on a wheat bran based medium enriched with Se could enhance the inorganic Se transformation improving the bioavailability of the mineral,and thus,the nutritional value of the wheat bran/mushroom powder.
Study on Periodic physiology of Phytoremediation for Formaldehyde

Abstract:
Through the determination of photosynthetic characteristics physiological indexes and formaldehyde content in leaves of Sansevieria thunb,Chlorophytum comosum,Chamaedorea elegans,Spathiphyllum floribundum and Hedera helix,the periodic physiology of phytoremediation for formaldehyde was analyzed and discussed.The result showed that: the diurnal variations of formaldehyde content in plants presented as that achieved the highest at about 8:00,then gradually decreased with the lapse of time,and reached the lowest at about 12:00,after gradually increased with the time expand.The relationship with photosynthetic characteristics manifested as that with the improving of Pn,Tr,Ci,Gs declined and the reducing ascended,with the reducing of Ls declined and the improving ascended,and the effect of WuE on the diurnal variations of formaldehyde content in plants was not obvious.With the changing of photosynthetic characteristics,the daily average content of formaldehyde demonstrated wave properties.The partial regression and correlation analysis showed that the formaldehyde content in leaves of Sansevieria thumb was related to Tr,Ls,Ci,with the partial regression very significant;the formaldehyde content in leaves of Chlorophytum comosum had relation to Pn,Ls,Ci of partial regression very significant,and Gs of significant;the formaldehyde content in leaves of Chamaedorea elegans was related to Pn,Gs,WuE,with the partial regression very significant,while Ls,Ci significant;the formaldehyde content in leaves of Spathiphyllum floribundum had relation to Tr,Pn,Gs of partial regression very significant,and Ci,WuE of significant;the formaldehyde content in leaves of Hedera helix was related to Tr,Ci,Gs,with the partial regression very significant,while Pn,Ls significant.It can be seen that the period of phytoremediation for formaldehyde was related to photosynthetic physiology of plants.

Abstract:
Many Bacillus-like species samples were collected by the Institute of Agricultural Bioresource,Fujian Academy of Agricultural Sciences(FAAS),and their classification characterization,statistics and the property of metabolism had been studied.In order to make a better management of Bacillus-like species sample database,a SQL Server database of Bacillus-like species samples was constructed,where the open database connectivity(ODBC) was used to the communications between clients and server on internet.All informations were stored in the QR code and a scanning gun can be used to do quick navigation of samples and rapid lookup of sample information.Bacillus-like species sample database could custom any sample information,customer could accomplish multi-conditions retrieval and statistics.
Techniques of Double Water Pressure Biogas Digesters Made of Glass Fiber Reinforced Plastic and its Application Benefit

Abstract:
This article introduces the structure and fermentation technology of the double water pressure biogas digesters(BHZJ) made of glass fiber reinforced plastic.Comparing to conventional digesters,results indicated that the biogas generating rate of BHZJ biogas digesters was 0.381 m3·m-3·d-1 in daily average per volume and the daily average gas production was 2.30 m3,which increased by 27.0% and 27.8%,respectively.To evaluate the benefit in use of the biogas digester,the data showed that the annual energy benefits of biogas production in coal-equivalent and firewood-equivalent was 2.613 t·a-1 and 2.51 t·a-1,respectively,which reduced the cost for energy consumption in daily life by 1 251.6 Yuan·a-1.The emission of CO2 reduced by using biogas substitute for energy.The reducing amount of CO2 emission in substitute for coal and firewood was by 3.608 8 t·a-1 and 3.585 4 t·a-1,respectively.
Studying on Root System of Wax Jambu,Syzygium samarangense in Morphology and Distribution

Abstract:
The varieties of Wax Jambu,Syzygium samarangense var.nongke 1 and var.nongke 2,were studied for observing in their root system morphology and distribution.The vertical depth of root system reached to 80 cm in the soil layer for both varieties.The root system of 'var.nongke 1' was mainly fastened in the 20-60 cm of the soil layer,while that of 'var.nongke 2' in the 0-40 cm.The diameter in single root measured was between 0-5 mm mostly the root system.The horizontal root area of the 6-year-old 'var.nongke 2' was in 4.56 m × 2.88 m.The distribution of active root tops was reached in 1.34-2.28 m away from the root collar.
