辽棉系列品种黄萎病抗性综合评价

Verticillium Wilt Resistance of Liaomian Cottons

    目的 综合评价辽棉系列棉花品种黄萎病抗性的现状和发展过程。
    方法 于2022~2023年,采用多种抗病鉴定方法对29个辽棉品种的抗病性进行鉴定,包括田间病圃鉴定、大丽轮枝菌Vd991室内接种后苗期抗病性鉴定、幼苗茎部活性氧(ROS)爆发染色鉴定。
    结果 通过比较各品种的病情指数发现:辽棉系列品种在辽宁辽阳和河北石家庄两地病圃都表现出了较强的抗病性,13个品种同时表现抗或高抗黄萎病;室内苗期鉴定结果病情指数偏高,有12个品种表现耐病,基本趋势与田间鉴定结果相似。活性氧(ROS)爆发后,采用NBT和DAB进行染色,有11个品种同时表现抗病。综合以上鉴定结果发现:LM19、LM23、LM25、LM27、LM33、LM40、LM42共7个品种抗病性表现稳定且突出。以两地田间鉴定结果和室内苗期鉴定结果、幼苗茎部活性氧(ROS)爆发DAB和NBT染色结果(含对照)为基础,分别采用标准化转换—欧式距离—离差平方和法进行聚类分析,在欧氏距离3.25和4.0处,将所有品种分别分成5个类群,比较各类群的分布及所包含品种的抗病性及育成时间发现,辽棉系列品种的抗病性随育成时间的延后而加强。
    结论 辽棉系列棉花品种具有极强的抗病潜力,可以作为资源材料在育种和生产实践中应用。通过资源创新不断获得对新生成病菌生理小种抗性的过程,可以为抗病棉花育种提供参考。

     

    Abstract:
    Objective The resistance of Liaomian cotton to verticillium wilt was studied to aid disease control.
    Methods In 2022-2023, 29 varieties of Liaomian cotton in Liaoning Province were evaluated for their resistance to verticillium wilt using multiple means including field disease nursery evaluation, in vitro Verticillium dahliae Vd991 inoculation on seedlings, and staining assay on burst of reactive oxygen species (ROS) in seedling stems and Morks as control. Euclidean distance-optimized data transformation with sum of squared deviations was applied to cluster the selected resistant species for classification.
    Results The Liaomian series of cotton cultivars displayed significant resistance to the wilt at the nurseries in Liaoyang, Liaoning and Shijiazhuang, Hebei. Among them, 13 varieties were rated resistant (R) or highly resistant (HR). Although the indices of the in vitro inoculation test on the seedlings were relatively high, 12 of them were shown to be tolerant of the disease, as similarly indicated in the field test. In addition, the NBT and DAB staining on the seedling stems after ROS burst suggested that 11 varieties to be wilt resistant. Overall, LM19, LM23, LM25, LM27, LM33, LM40, and LM42 displayed stable and considerate resistance to verticillium wilt. The cluster analysis using the Euclidean distances of 3.25 and 4.0 classified the 29 cotton varieties into 5 categories. A comparative analysis on the cluster distribution revealed that the disease resistance of the plants tended to be higher on the more recent bred varieties.
    Conclusion The varieties of Liaomian cotton carried verticillium wilt resistance in the genetic pool. Through enhancing the existing and/or incorporating new resources, they could be applied in breeding disease-resistant cotton for farming.
