响应面法优化猪肚菇液体菌种发酵配方

Response Surface-optimized Formulation for Culturing Pleurotus giganteus Spawns

    目的 为优化猪肚菇液体菌种培养基配方、制备优良液体菌种、推动其工厂化生产快速发展,本研究旨在采用响应面法,以菌丝生物量为指标,探究不同碳源、氮源、无机盐种类及添加量对猪肚菇液体菌种生长的影响,旨在筛选最适合其液体菌种生长培养基配方。
    方法 试验使用三角摇瓶培养方法,以猪肚菇菌株ZD9为供试材料,进行培养基各营养成分的单因素试验,系统分析6种碳源、8种氮源、11种无机盐对其菌丝生长的影响;在此基础上,采用Box-Behnken响应面法优化并验证猪肚菇液体菌种生产的培养基配方。
    结果 猪肚菇液体菌种发酵培养至第6天时,菌丝生物量达到峰值,菌丝干重为1.33 g·100 mL−1;适合其菌丝生长的营养要素为全麦粉、豆粕粉、MgSO4·7H2O及KCl,最优配方为全麦粉70 g·L−1、豆粕粉27 g·L−1、MgSO4·7H2O 2 g·L−1、KCl 2 g·L−1。该配方下的菌丝生物量实测值为2.51 g·100 mL−1
    结论 通过响应面法优化可获得适合猪肚菇液体菌种生长的培养基配方,经验证优化后的发酵配方较初始培养基菌丝生物量提高了89%,其菌丝生物量达到预期效果。

     

    Abstract:
    Objective Culture medium for growing Pleurotus giganteus spawns was formulated by means of response surface optimization.
    Methods  A single factor fermentation experiment was carried out in triangular shaking flasks to evaluate the effects of 6 carbon sources, 8 nitrogen sources, and 11 inorganic salts on mycelial growth of P. giganteus ZD9. Box-Behnken response surface design was used to optimize the medium formula.
    Results  The dry weight biomass of the spawns peaked at 1.33 g·100 mL−1 on the 6th day of culture in the optimized medium, which was formulated with the following ingredients at per liter rates of 70 g whole wheat flour, 27 g soybean meal powder, 2 g KCl, and 2 g MgSO4·7H2O to obtain a mycelial biomass at the concentration of 2.51 g·100 mL−1.
    Conclusion  The optimized medium for culturing P. giganteus spawns achieved an 89% improvement on mycelial biomass production.
