全国大蒜主产区蒜田内蝇类害虫种类调查及主要种类害虫共生菌比较研究

Pest Flies in Garlic-producing Regions of China and Symbiotic Microbes in Dominant Fly Species

    目的 系统厘清我国大蒜主产区蝇类害虫物种组成、区域分布特征,明确主要害虫田间快速形态鉴别特征,并解析优势害虫体内共生细菌、真菌群落差异,弥补现有调查仅单一关注葱地种蝇、缺乏全国尺度害虫本底与共生微生物基础数据的研究短板。
    方法 选取吉林、天津、山东、河南、江苏、湖北、云南7省共8处典型大蒜产区,采用扫网法采集蝇类成虫;结合显微形态观察与线粒体COI基因分子测序完成害虫物种鉴定;通过16SrRNA与ITS高通量测序技术解析两种优势蝇类害虫共生微生物多样性。
    结果 全国蒜田主产区共鉴定蝇类害虫8种,分属6科7属,蝇类群落存在显著地域分化,种植区周边水系、轮作模式会影响害虫种类,全国多产区两种近缘地种蝇葱地种蝇、灰地种蝇为优势物种;明确复眼、胸部粉被、翅亚前缘脉、中胫节鬃毛4项形态指标能够区分两种近缘地种蝇;两近缘优势种共生细菌群落组成差异显著,真菌群落结构相对保守,枝孢属为共有核心真菌。
    结论 本研究明确葱地种蝇与灰地种蝇为我国大蒜主产区主要蝇类害虫,阐明两种近缘蝇类害虫共4个形态鉴别要点及其共生菌组成差异:细菌群落受物种、性别共同调控;真菌群落中枝孢属为共有核心真菌。这为蒜田蝇类精准监测及后续害虫共生菌机制研究提供理论依据。

     

    Abstract:
    Objective Species and distribution of pest flies found in the major garlic-producing areas of China were identified, and endosymbiotic microbes in the dominant fly species compared for improved pest control.
    Method At 8 typical garlic fields in 7 provinces in China including Jilin, Tianjin, Shandong, Henan, Jiangsu, Hubei, and Yunnan, adult flies were captured by sweep-netting for the study. Species of the caught pests were determined by microscopic morphological observation and mitochondrial COI gene sequencing with dominant species identified. Symbiotic microorganisms in the top two dominant species were classified by high-throughput sequencing of 16S rRNA and ITS genes.
    Result There were 8 species belonging to 7 genera of 6 families identified in the total collected specimens. Significant diversity was found in the populations of different localities, fields how close to water or practicing varied crop rotation. In the regions, congeneric Delia antiqua and D. platura were the dominant species of pest flies. They had distinctively different compound eyes, thoracic pruinose coating, wing subcostal veins, and mid-tibial bristles. The symbiotic microbial communities in them significantly differentiated, with the fungi being relatively conserved and Cladosporium the common genus.
    Conclusion D. antiqua and D. platura were identified as the primary pest fly species found in the garlic fields in the regions. Although closely related, they could be reliably distinguished by morphological characteristics. Depending upon the species and sex as well as the field condition where these flies inhibited, their symbiotic microbial communities varied significantly in composition but shared Cladosporium as the common fungal taxon.
