xatB基因簇在水稻细菌性条斑病菌致病力调控中的作用

Regulatory Function of xatB Gene Cluster in Virulence of Xanthomonas oryzae pv. oryzicola

    目的 明确xatB基因簇在水稻细菌性条斑病菌(Xanthomonas oryzae pv. oryzicolaXoc)致病过程中的调控作用。
    方法 构建该基因簇中3个基因xatBXoc_1597mfd)和Xoc_1599rlmJ)的单突、双突和三重突变体及其互补菌株;打孔法检测这些菌株的致病力;通过平板试验比较野生型菌株与这些突变菌株和互补菌株的生长、胞外酶活性、运动能力及致病力等的差异。
    结果 xatBmfdrlmJ突变均导致Xoc致病力显著下降。rlmJ突变抑制Xoc生长、运动力和胞外多糖合成,而mfd突变则促进Xoc生长、运动力和胞外多糖合成,mfd互补则抑制细菌的生长、运动力和胞外多糖合成。双突和三突突变体与单突的表型不一致,呈现出拮抗或协同的作用。
    结论 以上结果表明,XatB、Mfd和RlmJ均是Xoc致病力的重要调节因子,三者之间呈现复杂的促进或抑制关系,具体机制还有待进一步研究。

     

    Abstract:
    Objective Role of xatB gene cluster played in regulating the infection of Xanthomonas oryzae pv. oryzacola (Xoc) on rice plants was investigated.
    Method Single, double, and triple mutants of Xoc on xatB, Xoc_1597 (mfd), and Xoc_1599 (rlmJ) in the cluster as well as the corresponding complemented strains were constructed. Pathogenicity of the mutated and complemented wild-type strains were detected by a punching method, and growth, extracellular enzyme activities, and motility determined by plate culture.
    Result All xatB, mfd, and rlmJ mutations resulted in a significant decline in the virulence of Xoc. However, the rlmJ mutation inhibited, while mfd promoted, the growth, motility, and extracellular polysaccharide (EPS) synthesis; but the mfd complementation retarded those functions of the pathogen. The double and triple mutants differed from the single mutants in phenotypes as well as antagonistic or synergistic effects on Xoc.
    Conclusion XatB, Mfd, and RlmJ significantly affected the Xoc virulence on rice plants. The complex relationship requires further investigation.
