甜玉米闽甜6855籽粒中维生素E和类胡萝卜素合成基因挖掘与分析

    目的 通过转录组挖掘潜在的合成调控基因分析甜玉米籽粒中维生素E和类胡萝卜素含量变化。
    方法 以‘闽甜6855’品种作为试验材料,利用分光光度法测定籽粒中维生素E和类胡萝卜素含量;利用转录组进行Mfuzz聚类分析、GO和KEGG分析,挖掘潜在的候选基因。
    结果 闽甜6855籽粒中维生素E含量从授粉后10 d至15 d先下降,随后逐渐增加;类胡萝卜素含量随籽粒发育成熟呈现显著增加的趋势。Mfuzz聚类分析获得6个随时间表达变化的基因集群,其中Cluster 6基因集表达趋势与维生素E和类胡萝卜素含量变化趋势相似。GO和KEGG富集分析发现,Cluster 6基因集能同时富集到维生素生物代谢和类胡萝卜素生物合成过程。对60个潜在维生素E调控基因和40个类胡萝卜素合成相关基因进行表达分析,发现Zm00001d005351Zm00001d052247Zm00001d015356Zm00001d016441Zm00001d045610表达与维生素E含量变化趋势相似;Zm00001d038170Zm00001d036345Zm00001d007502Zm00001d018869表达与类胡萝卜素含量变化趋势相似。
    结论 本研究通过对鲜籽粒中维生素E和类胡萝卜素含量测定与转录组分析相结合,鉴定出5个潜在调控维生素E生物合成基因和4个类胡萝卜素生物合成调控基因,为鲜食玉米VE和VA营养元素育种提供潜在的基因资源。

     

    Abstract:
    Objective Contents of vitamin E (VE) and carotenoid in developing sweet maize kernels were correlated to genes associated with their syntheses to aid breeding new varieties.
    Method VE and carotenoid contents in the kernels of Mintian 6855 were determined using spectrophotometry. Transcriptomes of candidate genes were used to identify those related to the formation of the nutrients by Mfuzz clustering, GO, and KEGG analyses.
    Result In the kernels, VE decreased in 10-15d after pollination and gradually increased afterward, while carotenoid continuously increased from the beginning till maturation. The Mfuzz clustering analysis identified 6 gene clusters with varied temporal expression patterns. Among them, Cluster 6 expressed in trends significantly paralleled the changing patterns of VE and carotenoid. It simultaneously enriched vitamin metabolic and carotenoid biosynthetic processes, as indicated by GO and KEGG. Of the 60 genes involved in VE regulation and 40 associated with carotenoid synthesis, Zm00001d005351, Zm00001d052247, Zm00001d015356, Zm00001d016441, and Zm00001d045610 expressed in a trend similar to that of changing VE, while Zm00001d038170, Zm00001d036345, Zm00001d007502, and Zm00001d018869 close to that of carotenoid increase in the kernels.
    Conclusion Five genes involved in regulating VE biosynthesis and 4 associated with carotenoid formation in kernels of Mintian 6855 were identified by this study.
